Formation of Adventitious Bud from tulipa gesneriana ¢¥ Red Matador ¢¥ Callus





Abstract
This study was carried out to establish the new method of micropropagaio of Tulipa gesneriana `Red Matador¢¥. Callus has been induced from the excised scale or floral stem explants on Murashige and Skoog¢¥s (MS) solid media with growth regulators. In scale explants, callus formation was enhanced in the dark cultural condition, on the bulb desication treatment for 10 days at room temperature, and the callus was observed at about 70 days after primary culture. In floral stem explants, callus formation was enhanced by 16 hr day lengh or by 10 §·/§¤ NAA, and callus forming was observed at 56.6 days after primary culture. Callus forming rate was 70% in both the scale and floral stem explants. Callus proliferation and adventitious buds have been induced successfully at MS solid medium containing 0.05§·/§¤ NAA and 1.0§·/§¤ BA by culturing callus, which have been derived from the scale or floral stem explants.
